Sephiroth's Intervention targets a creature when cast; if that target becomes illegal before the spell resolves (for example, by gaining protection from the spell's controller or no longer being on the battlefield), the spell is countered on resolution and nothing happens — you don't destroy a creature and you don't gain life.
If the targeted creature is destroyed by the spell, it goes to the appropriate zone (usually the graveyard) and then you gain 2 life as part of the same spell's effects resolving.
If replacement effects or replacement-duration abilities (for example, "If a creature would be destroyed, exile it instead") apply to the creature, those effects happen instead of being destroyed; the spell still resolves, but whether you gain life depends on whether the spell 'resolves' successfully—if the destruction is replaced but the spell still resolves, you do gain 2 life. (Note: based only on the provided oracle and ruling, the key point is that if the spell doesn't resolve because the target is illegal, you don't gain life.)
