If you cast Vault Skyward after blockers are declared, it won't make a creature that's already blocked become unblocked — giving flying then doesn't remove existing blocks.
You may target any creature, including one that already has flying or is already untapped; the spell still resolves and applies both effects.
If the targeted creature becomes an illegal target before resolution (e.g., it gains shroud or leaves the battlefield), Vault Skyward is countered by game rules and has no effect.

For flying to work as an evasion ability, Vault Skyward must be cast before the declare blockers step begins. Once a creature has become blocked, giving it flying won’t change that.
